Abstract

The goal of this project was to design and construct the setup incorporating one of the super resolution optical approaches Stimulated Emission Depletion microscopy or STED into our SFCS method with the ultimate goal to characterise the chromatin structure with sub diffraction precision This theses thereby describes the design implementation and calibration of a STED SFCS setup reaching a resolution of 60 nm Further technical improvements will allow us to improve the resolution down to 30 nm
